The point is, if they have a record that you are willing to pay and that you have properly sent the check, I don't think the clinic will impose any penalty on you.
If you called, please note the date and time of the call and the name of the person who answered the phone so that if you do not receive the mail, you can prove that you intended to pay and that you checked with the clinic.

I am not a fan of paying by check mail either.
If you receive a bill and the clinic does not have an online credit card payment portal, you can call the clinic directly and ask if you can verbally pay by credit card over the phone ？ and they will accept the card payment over the phone. Some clinics will accept credit card payment over the phone.
